Retųjų žemės elementų druskų poveikio vienaląsčiams dumbliams Pseudokirchneriella subcapitata lyginamasis tyrimas taikant tėkmės citometriją ir spektrofotometriją
Mikrodumblių augimui įtaką daro metalų jonai, kurie reguliuoja ląstelės biologinius procesus. Magistrinio darbo metu buvo siekiama nustatyti neesminių elementų (lantanoidų) įtaką Pseudokirchneriella subcapitata augimui per 72 valandas. Pasirinkti keturi retųjų žemių elementų: lantano, lutecio, gadolinio ir itrio chloridai. Ląstelių augimo slopinimas buvo vertintas pasitelkiant du metodus: tėkmės citometriją ir spektrofotometriją. Nustatyti tirtųjų lantanoidų veikliųjų koncentracijų diapazonai (3,15 - 4,27 mg/l tėkmės citometru ir 0,99 - 3,76 mg/l spektrofotometru), kuriuose mikrodumblių augimas buvo slopinamas 50 %. Gauti duomenys leidžia manyti, kad atskleidžiant atskirų lantanoidų poveikio ir jo dinamikos aspektus didesne skiriamąja geba pasižymi tekmės citometrijos metodas.
The growth of microalgae is influenced by metal ions, which regulate the biological processes of the cell. The aim of the master's thesis was to determine the influence of non-essential elements (lanthanides) on the growth of Pseudokirchneriella subcapitata in 72 hours. Four rare earth elements were selected: lanthanum, lutecium, gadolinium and yttrium chlorides. Inhibition of cell growth was assessed by two methods: flow cytometry and spectrophotometry. The ranges of active concentrations of the tested lanthanides (3.15 - 4.27 mg / l with a flow cytometer and 0.99 - 3.76 mg / l with a spectrophotometer) were determined, in which the growth of microalgae was inhibited by 50%. The obtained data suggest that the flow cytometry method has a higher resolution in revealing the aspects of the effect of lanthanides and its dynamics.
