Gyvų ląstelių mechaninių savybių tyrimas

Baigiamajame magistro darbe nagrinėjamas gyvų ląstelių mechaninių savybių nustatymas taikant atominių jėgų mikroskopą (AJM) bei matematinį modelį. Tiriamos Saccharomyces cerevisiae mielių ląstelės, pasižyminčios panašiomis savybėmis kaip ir žinduolių ląstelės. Siekiant ištirti ląstelių mechaninių savybių pokyčius, jos buvo inkubuojamos 9 µM bei 27 µM koncentracijos 9,10-fenantrenchinono (PQ) tirpale. Analizuojant rezultatus, gauta, kad šis tirpalas daro įtaką mielių ląstelių sienelių mechaninėms savybėms. Inkubuojant mieles 9 μM koncentracijos PQ tirpale medžiagos šiurkštumas ir tamprumo modulis beveik nesiskiria nuo kontrolinės grupės rezultatų. Tačiau lyginant kontrolinę grupę su 27 μM PQ koncentracijos paveiktomis ląstelėmis - tamprumo modulis 2 kartus didesnis, o šiurkštumas kito nuo 2,53 iki 3,84 nm. Sukurtas mielės ląstelės baigtinių elementų modelis, kuris buvo taikomas nustatyti mielių sienelės, membranos bei citoplazmos struktūrų mechaninių savybių įtaką eksperimentiniu būdu gautoms ląstelių mechaninėms savybėms.Darbo apimtis - 51 p. be priedų, 45 iliustracijų, 3 lentelės, 33 literatūros šaltiniai. The final master's thesis deals with the determination of the mechanical properties of living cells using atomic force microscope (AFM) and a mathematical model. Saccharomyces cerevisiae yeast cells are studied, which have similar properties to mammalian cells. In order to determine the changes in the mechanical properties of the cells, they were incubated in 9 µM and 27 µM 9,10-phenanthrenquinone (PQ) solution. Analysis of the results showed that this solution affects the mechanical properties of the yeast cell walls. When incubating yeast in 9 μM PQ solution, the roughness and the elasticity modulus of the material are almost the same as in the control group. However, when comparing the control group with the cells affected by the PQ concentration of 27 μM, the modulus of elasticity was 2 times higher and the roughness varied from 2.53 to 3.84 nm. A model of yeast cell finite elements was developed, which was applied to determine the influence of the mechanical properties of the yeast wall, membrane and cytoplasmic structures on the experimentally obtained mechanical properties of the cells.Thesis consists of: 51 p. without appendixes, 45 illustrations, 3 tables, 33 references.
