Egzosomų savybių tyrimas in vitro bei in vivo, vertinant galimybę veikti onkologinio proceso eigą
Exosomes - nano-sized particles that are actively secreted by cells. It is an active highly stable membrane cell communication tool that contains MHC Class I and II complexes, costimulatory molecules, chaperone, tetraspanins, various adhesives, and unique molecules for each cell type. Cancer cell exosomes concentrate information on cancer antigens, so exosomes could potentially be used in immunotherapy. Exosomes of different cancer cell lines were characterized in this work. To assess whether exosomes can activate immune cells, dendritic cell maturation under the influence of cancer exosomes was observed. Dendritic cells that were matured with one of the selected cancer exosomes were tested in an in vivo mouse model. Finally, it was determined whether the exosomes of the immune cells acting on immunogens could affect the proliferation of other leukocytes. During the work it was found that the exosomes of different cancer cells differ in size, and the amount of exosomes isolated depends not only on the number of cells, but also on their type. It has also been observed that glioblastoma GL261 exosomes differ in their marker profile from other cancer cells. It has been shown that the exosomes of all cancer cells activate the maturation of dendritic cells, but no differences were found between dendritic cell vaccines in treated tumor mice. Nevertheless, it has been shown that stress-experienced leukocytes produce exosomes that activate leukocytes that have not been exposed to immunogens.
