Sidabro nanodalelių poveikis mielių saccharomyces cerevisiae ląstelėms
Šio darbo tikslas - ištirti sidabro nanodalelių toksiškumo įtaką mielių S.cerevisiae ląstelių kultūrai in vitro. Darbo apimtis - 76 p. teksto be priedų, 49 iliustr., 6 lent., 74 bibliografiniai šaltiniai.
The aim of this work was to identify the toxicity of the silver nanoparticles for the S.cerevisiae yeast cells in vitro. Silver nanoparticles (Ag Np) were synthesized by chemical synthesis using Lee-Meisel and Creighton methods. By using UV-Vis spectroscopy, atomic force microscopy (AFM) and transmission electron microscopy (TEM), it was found that size of Ag Np synthesized by Lee-Meisel is 4 to 80 nm, their shape was spherical and their surface Plasmon resonance (SPR) was at 428 nm. Ag Np obtained, by Creighton method, size is from 5 to 160 nm, their shape is spherical and their SPR is at 398 nm. Also, these methods have been tested on Ag Np, that are commercially available Ag Np with the size of 10-150 nm, spherical shape and with SPR at 411 nm. Yeast cells that were grown in liquid YEPD medium, together with silver nanoparticles and AgNO3, were investigated by AFM and Raman spectroscopy. The results obtained by AFM showed that the major changes were observed in yeast cells that were exposed to by Lee-Meisel method synthesized Ag Np, also the biggest impact on yeast cell wall elasticity had by Lee-Meisel synthesized Ag Np. Raman spectroscopy showed molecular changes in yeast cells, which indicate the destruction of cell wall components. The largest molecular changes seen in yeast cells exposed to by Lee-Meisel and Creighton obtained synthesis silver nanoparticles. Also, yeast cells exposed to Ag Np were deposited on an agar medium, where the effect of nanoparticles on yeast colony formation was determined. This has shown that Ag Np synthesized by Lee-Meisel and Creighton methods also have biggest impact on yeast growth.Structure: introduction, literature review, materials and methods, results and discussion, conclusions and references.Thesis consists of: 76 p. of text without appendixes, 49 pictures, 6 tables, and 74 bibliographical sources.
