Mikrodalelių atranka bei atskyrimas iš mišinių - fundamentali biologinių tyrimų dalis. Dauguma atrankos sistemų turi apribojimus, kurie lemia ribotą jų taikymą tyrimuose - priklausomos nuo antikūnų sąveikos, apribotos fluorescencijos analize ar pasižymi mažu našumu. Baigiamojo darbo metu sukurta fotopolimerizacija paremta mikrodalelių atrankos sistema - patobulinus įvairius esamus mikroskopijos metodus, tačiau išlaikant didelį atrankos našumą, kuriuo pasižymi FACS. Optimizavus fotopolimerizacijos metodiką, įvertintas atrankos sistemos efektyvumas, greitis bei tikslumas ir ji pritaikyta tikslinių mikrokapsulių išgryninimui. Atliktas optimalios lazerio trukmės ir polimero koncentracijos įvertinimas - C (Gel-MA) = 4,5% ir t (lazerio) = 50-100 ms. Įvertintas lazerio taško dydis bei hidrogelio plėtra - vidinių bei išorinių hidrogelių plotų diametrai didėja keliant LAP koncentraciją. Atliktas hidrogelio skaidymo greičio įvertinimas - skaidymas vidutiniškai trunka 5,8 s, naudojant 10 mg/ml kolagenazės tirpalą. Paruoštos fluorescuojančios mikrokapsulės atrankos sistemos įvertinimui. Pademonstruota, jog atrankos sistema gali būti pritaikyta tikslinių mikrokapsulių išgryninimui - hidrogeliuose užlaikytų mikrokapsulių atpalaidavimas trunka iki 10 s, naudojant 1 mg/ml kolagenazės tirpalą. Įvertintas atrankos sistemos efektyvumo vidurkis x̄ = 85,39%, standartinė paklaida σ = 2,22 % (vidutiniškai 85,39% visų užlaikytų mikrokapsulių populiacijos sudaro identifikuotos) bei tikslumo vidurkis x̄ = 93,74%, standartinė paklaida σ = 2,81 % (atrankos sistema vidutiniškai identifikuoja 93,74% visų fluorescuojančių mikrokapsulių). Išmatuota, jog vienas atrankos sistemos ciklas trunka 8,3 s. Atrankos sistema identifikuoja ne visą fluorescuojančių mikrokapsulių populiacijos dalį bei užlaiko ne tik identifikuotas, bet ir kitas šalia esančias mikrokapsules.Darbą sudaro septynios dalys: įvadas, literatūros apžvalga, medžiagos ir metodai, rezultatai ir jų aptarimas, išvados, literatūros šaltiniai, padėka. Screening and separation of microparticles is a fundamental part of biological research. Most systems have limitations that result in their limited application in research - they depend on antibody interactions, are constrained by fluorescence analysis, or have low-throughput. During the thesis, a microparticle screening system based on photopolymerization was developed. The system improves existing microscopy methods while maintaining high-throughput characteristics of FACS. After optimizing the photopolymerization methodology, the specificity, throughput, and sensitivity of the screening system were evaluated, and it was adapted for the separation of microcapsules. An evaluation of the optimal laser duration and polymer concentration was performed - C (Gel-MA) = 4,5% and t (laser) = 50 - 100 ms. Laser spot size and hydrogel expansion were evaluated - the internal and external diameters of the hydrogels increase with rising LAP concentration. The digestion rate of the hydrogel was evaluated - it takes 5,8 seconds on average when using a 10 mg/ml collagenase solution. Fluorescent microcapsules were prepared for the evaluation of the screening system. It was demonstrated that the system can be adapted for the separation of target microcapsules - digestion of hydrogels takes up to 10 seconds when using a 1 mg/ml collagenase solution. Average specificity of the screening system was evaluated to be x̄ = 85,39% (standard error σ = 2,22 %) (on average, 85,39% of all retained microcapsules were identified), and the average sensitivity x̄ = 93,74% (standard error σ = 2,81 %) (the system identifies 93,74% of all fluorescent microcapsules). It was measured that one cycle of the system takes 8,3 seconds. The system does not identify the entire population of fluorescent microcapsules and retains not only the identified ones, but also other nearby ones.Structure: introduction, literature review, methods and materials, results and discussion, conclusions, references, and acknowledgment.
