Investigation of the Role of Neuronal Interleukin 16 in Chondrogenic Differentiation
The objective of this study was to elucidate the role of neuronal interleukin 16 (NIL-16) in chondrogenic differentiation of human mesenchymal stem cells (hMSCs). To achieve this objective the expression of NIL-16 and transient receptor potential vanilloid 4 (TRPV4) in chondrogenic differentiation of human and mouse mesenchymal stem cells in osteoarthritic cartilage tissue was determined to establish whether NIL-16 molecule acts to stabilise ion channels in osteoarthritic chondrocytes. In order to determine the expression of NIL-16 and confirm whether it is involved in hMSC chondrogenesis, detection of the protein was carried out. Levels of NIL-16 protein expression were high from Day 0 and gradually decreased until Day 21 of chondrogenesis. The experimental evidence from the cell surface staining showed that NIL-16 might be seen at the cell surface of hypertrophic cells as it is only seen in hypertrophic cells and osteoarthritis (OA) tissue. Moreover, the pattern of cell surface staining of NIL-16 in hypertrophic chondrocytes has been provided; it showed that NIL-16 co-localises with TRVP4. In this study, for the first time NIL-16 has been associated with chondrogenic differentiation in MSCs by binding to TRPV4 and stabilizing ion channels and it might be involved in establishing the hypertrophic OA chondrocyte in late OA.
