The purpose of this final work is to investigate the interaction of different metal ions with recombinant porcine, mink, human growth hormones. Cations which influence on the oligomeric structures of recombinant human, porcine, mink growth hormones in water are known were chosen. Using thermal shift assay it is determined that Zn2+, Cu2+, Ni2+, Cd2+, Co2+ destabilize recombinant porcine and mink growth hormones. A quantitative model of protein thermal stabilization and destabilization by ligand was used to determine these metal ions binding constants to the unfolded states of the rpGH, rmGH. Conversely Zn2+, Cd2+, Ni2+, Co2+ stabilize recombinant human growth hormone but there are critical concentrations of metal ions. When the Zn2+, Cd2+, Ni2+, Co2+ concentrations exceed their critical concentrations these metal ions destabilize rhGH. Mn2+ do not interact with recombinant porcine, mink growth hormones. Mn2+ and Cu2+ do not effect the stability of rhGH, but bigger concentrations of these two metal ions also destabilize recombinant human growth hormone. Also stoichiometry, constants and enthalpies of Zn2+ binding to native rpGH and rmGH are determined directly by isothermal titration calorimetry. Additionally ITC experiments confirm that one zinc ion binds in a cooperative manner to one recombinant human growth hormone molecule.
